RESUMO

Objective:To investigate the effectiveness of nasal endoscopic anterior lacrimal recess approach combined with temporary fenestration of the nasal septum in resecting recurrent nasal inverted papilloma. Methods:Patients with recurrent nasal inverted papilloma who underwent reoperation in our hospital during the past 2 years were included . The nasal septum may hinder full access to and effective treatment of the lesions at the anterior and medial wall of the maxillary sinus by endoscope, aspirator and surgical instrument in the narrow aperture of the prelacrimal recess, although these lesions could be observed by 70° nasal endoscope. Results:The nasal septum is temporarily opened on the basis of the prelacrimal recess approach, and the nasal endoscope and instrument was introduced through trans-septal window, so as to provide a better view of the operative field and the angular range of the instrument's movement. Conclusion:The recurrent nasal inverted papilloma could be successfully managed by re-endoscopic anterior lacrimal recess approach combined with temporary fenestration of the nasal septum, and no recurrence was observed during the 2-year follow-up. This surgical approach is recommended for the inverted papilla which originates from the anterior medial wall of the maxillary sinus, as the tumor can be removed completely using this surgical approach.

OBJECTIVE To investigate the effects of behavior, pathology, the serum IL-17, IL-23 level, and the expressing of RORγt, IL-17 and IL-23 mRNA in nasal tissues of experimental allergic rhinitis rats after the scoparone treatment. METHODS The animal model were divided into 4 groups: normal control group(group NC), allergic rhinitis group(group AR), artemolactone group(group Sco) and dexamethasone group(group Dxm). The symptom score, HE staining of the nasal mucosa, IL-17 and IL-23 level in serum measured by ELISA, the RORγt, IL-17 and IL-23 mRNA level detected by RTPCR. RESULTS Symptoms and inflammatory pathology were relieved in the experimental group after scoparone treatment. The serum levels of the IL-17, IL-23 in group Sco and group Dxm were little higher than that in group NC. The levels of RORγt, IL-17 and IL-23 mRNA in group AR were significantly higher than that in the other three groups. The levels of RORγt, IL-17 and IL-23 mRNA in group Sco and group Dxm were little higher than that in the group NC. CONCLUSION Sco could significantly inhibited or eliminated the allergy symptoms of AR in rats, and could reduce the severity and inflammatory response of diseases.

OBJECTIVE@#To test the immunoglobulin free light chain (FLC) from nasal secretion and serum of patients with allergic rhinitis(AR)and non-allergic rhinitis(NAR) for the purpose of exploring the possible immunological mechanism.@*METHOD@#Ninety consecutive patients were selected between January 2009 and January 2012, involving 45 patients with AR and 45 patients with NAR diagnosed by symptoms,signs,skin prick tests(SPT) and specific IgE (slgE). Forty-five volunteers were chosen as healthy control (HC). According to the visual analogue scale (VAS) scores,the nasal symptoms of AR and NAR,including sneeze. Nasal discharge. Nasal obstruction and nasal itching were compared. ELISA was used to detect the total IgE, IL-16, IL-17 in nasalsecretion and serum. The data was analyzed by SPSS 17.0 software.@*RESULT@#There was no statistical difference between AR and NAR group in nasal symptoms (P > 0.05); In serum, IL-16 and IL-17 increased in AR group comparared to NAR group (P < 0.05); IL-16 and IL-17 increased in NAR group comparared to HC group (all P < 0.05); In nasal secretion, IL-16 and IL-17 increased in NAR and AR group comparared to HC group (all P < 0.05).@*CONCLUSION@#IL-16, IL-17 takes part in the path of physiological process of AR and NAR with the immunological mechanism.

Objective To explore the effect of mouse proliferation-associated protein 2G4 (p38-2G4) high-expression on the proliferation and erythriod differentiation of murine erythroleukemia ( MEL ) cells.Methods To establish the recombinant lentivirus vector p 38-2G4-pLJM1, the p38-2G4-pLJM1 was cotransfected into HEK293T cells to obtain lentivirus with pCMV-VSV-G and pCMV-dR8.2.Lentivirus were infected into MEL cells to establish the stably p 38-2G4 high-expressed MEL cells.Western blotting was used to analyse the high-expression efficiency.MTT assay and benzidine staining were applied to detect the cell viability and hemoglobin synthesis of the stable cell line in presence /absence of inducers.Results Western blotting showed that the p38-2G4 high-expression stable cell strain had a higher expression of p38-2G4 as compared to the control group ( MEL) ( P 0.05), but the hemoglobin synthesis had been reduced as compared to the control group (P<0.05).Conclusion p38-2G4 high-expression does not affect the cell viability of MEL cells , but inhibits the erythriod differentiation of MEL cells in three independent experiments .

Objective To study NonO protein expression changes in murine erythroleukemia ( MEL ) cell differentiation induced by sodium butyrate .Methods Benzidine staining was used to test sodium butyrate-induced erythroid differentiation of MEL cells .We detected NonO protein expression changes in MEL cell differentiation induced by sodium butyrate and NonO protein localization in MEL cells by Western blotting and immunocytochemistry .Furthermore , we applied PCR technique to detect NonO RNA expression in differentiation process .Results We found that NonO protein was upregulated at gene and protein levels in the erythroid differentiation process of MEL cells induced by sodium butyrate and located in cytoplasm and nucleus in MEL cells .Conclusion These results show that NonO protein is closely related with MEL cell differentiation induced by sodium butyrate , which may provide important clues for further study of the mechanism of leukemia .

Objective To explore the differentially expressed proteins during erythroid differentiation .Methods The murine erythroleukemia ( MEL) cell were treated by DMSO , and the comparative proteomic was systematically analyzed and identified on different differentiating time points .ratio of cell differentiation and viability were detected by benzidine staining, MTT assay and Ter119 immunofluorescence.Using two-dimensional gel electrophoresis combined with mass spectrometry technology and bioinformatics analysis , we conducted a comparative proteomic analysis on MEL cells during the process of induced differentiation to screen and identify differential proteins .Results The MEL cells induced by 1.2%DMSO for 0 hour, 6hours, 12hours, 24hours, 36hours, 48hours, 72 hours, 96 hours, 120 hours were collected for proteomic analysis, by two-dimensional gel electrophoresis combined with mass spectrometry .A total of 87 kinds of proteins were successfully identified .MEL cells exposed to DMSO at a final concentration of 1.2% for 120 hours reached the highest differentiation rate of 67%.MTT assay showed that 1.0%, 1.2%, 1.4% DMSO had no inhibiting effect on cell vitality.Conclusion DMSO may induce MEL cells to differentiate and have no inhibiting effect on cell vitality .The 87 kinds of differentially expressed proteins from two-dimentional gel electrophoresis may be divided into twelve categories ;the most three parts are 41%enzyme protein, 15%structural protein and 13%regulatory protein.

RESUMO

RORγt, an immune cell-specific isoform of RORγ( retinoic acid receptor-related orphan nuclear receptor gam-ma) , is a key transcription factor for the development of Th 17 cells both in human and mouse .RORγt is required for the induction of IL-17 transcription and for the manifestation of Th 17-dependent autoimmune diseases in mice .RORγt natural ligands are retinoic acid . RORγt is closely implicated in the pathology of numerous autoimmune diseases , infectious diseases and cancer .With the further re-searches about the role of RORγt, we will clarify the mechanism of RORγt in autoimmune diseases , which will provide new ideas to di-agnose and treat autoimmune diseases .

OBJECTIVE@#To observe changes of serum Th1/Th2 cytokines and IgE of rats with experimental allergic rhinitis (AR) treating with Scoparone (20 ml/kg, the concentration is 168 mg/L).@*METHOD@#Forty male SD rats were randomly divided into 4 groups : group NC, group AR, group Sco, group Dxm. The rats were sensitized with OVA and were then treated with Scoparone, during the treatment, the behaviors were observed and the change in nasal mucosa were recorded. The level of serum cytokines IFN-gamma, IL-4, IL-5 and IgE were determined by ELISA.@*RESULTS@#The behavior scores of group Sco were significantly lower than those of group AR, the difference was statistically significant (P 0.05). The nasal inflammation of group Sco was significantly less than that of group AR. The level of serum IFN-gamma of group Sco was significantly higher than that of group AR, while the levels of serum IgE, IL4, IL-5 of group Sco were significantly lower (P 0.05).@*CONCLUSION@#Scoparone is highly effective in treating allergic rhinitis by regulating the expression of Th1/Th2 cytokines and IgE.

OBJECTIVE@#To observe the suppressive effects of immunostimulatory DNA sequences (ISS) in conjunction with dermato phalloides farinae allergen (Df) on nasal cavity inflammation in rats of experimental allergic rhinitis (AR).@*METHOD@#Thirty-two rats were divided into 4 groups: group ISS(A), group ISS+ Df (B), group Df (C) and group normal saline (D). Rats in groups A,B and C were sensitized and challenged with Df. Blood samples were obtained every week for six weeks, Df specific IgE was measured by ELISA. The nasal mucosa were studied pathologically. The levels of serum interferon gamma (IFN-gamma), IL-12, IL-4 and IL-5 were determined by ELISA.@*RESULT@#The serum level of sIgE in group B was significantly lower than that in group C in six weeks, but that in group A was not significantly different to that in group C. The mean levels of serum IFN-gamma and IL-12 in A, B group were significantly higher than in C group (P < 0.01). But IL-4 and IL-5 level was much lower (P < 0.01).@*CONCLUSION@#ISS+ Df have inhibited effectiveness on allergic nasal cavity inflammation in rats and its action time is 6 weeks.

<p><b>OBJECTIVE</b>Searching for more potent and less toxic HMBA-related agents.</p><p><b>METHODS</b>Human erythroleukemia cell K562, murine erythroleukemia cell (MEL) and its sub-line MEL DS19 were used as target cells to select a cell line which is the most sensitive to HMBA, then analyzed the activity of inducing differentiation of two new designed HMBA derivatives: HMBPA [hexamethylenebi (3-pyridin) amide] and Co-HDTA (ethylenediaminetetra acetic acid cobalt) using cell biology, cytochemical and molecular biology techniques.</p><p><b>RESULTS</b>We found that the MEL DS19 cells were most sensitive to HMBA (benzidine positive, B+ approximately 76%). Co-HDTA can inhibit the growth of MEL DS19, but induces differentiation just in a small population (B+ 2% approximately 4.5%). Between 0.02 approximately 5 micromol/L, HMBPA induces 3% approximately 8% cells committed to differentiation with little inhibition of cell proliferation. 1 micromol/L HMBPA and 2 mmol/L HMBA together, can obviously increase the percentage of differentiated cell (B+ approximately 72%), inhibit DNA synthesis and accelerate beta-globin transcription.</p><p><b>CONCLUSION</b>The new HMBA derivatives may provide potential cancer differentiation inducers.</p>

Objective:To observe the effects of sera Th1/Th2-typed cytokines in patients allergic rhinitis (AR) who were treated with kebimin decoction(KD),and to study the therapeutic mechanism of KD in AR.Methods:60 patients of AR were randomly divided into two groups (treating group,TG and control group,CG). Patients in TG were treated with KD,while those in CG were treated with Xinfang Rhinitis Capsule before and after treatment,the levels of serum Th1-typed cytokines IFN-?、IL-2、IL-12 and Th2-typed cytokines IL-4、IL-5、IL-10 were determined by enzyme-linked immune sorbent assay (ELISA); and compared with 30 healthy controls.Results:The levels of serum Th2-typed cytokines IL-4、IL-5、IL-10 were higher and the levels of Th1-typed cytokines IFN-?、IL-2、IL-12 were lower in AR than that in healthy control ( P 05).Conclusion:Kebimin decoction is highly effective in treating allergic rhinitis by regulating the levels of Th1、Th2-typed cytokines,correcting the imbalance Th1/Th2-typed cytokines network.

RESUMO

Objective To investigate the effect of optimized conditions on proliferation and differentiation of the neural stem cells(NSCs) from different age brain of rat in vitro. Methods The brain tissues from embryonic(E12~E15d),newborn(1~2?d) hippocampus and olfactory bulb from adult of rat were isolated.After proliferation in conditional culture medium including growth factor and induced differentiation,the cell type specific antibodies were used for the immune fluorescence staining to identify the cells. Results A lager scale NSCs were obtained from brain tissue and could proliferate in conditional culture in vitro. They could self renew and generate both nerve and glia.Conclusion\ The NSCs got from rat brain can be used for basic and clinical research.\;[

The synthesis of cellular and nuclear basic proteins in the first generation of the repopulated pachytene spermatocytes (RPS) after the withdrawal of gossypol treatment and the normal control pachytene spermatocyte (CPS) were studied by the incorporation of ~3H-arginine into proteins. After treatment with gossypol for various periods of time, the synthetic activity for cellular protein in the RPS was slightly inhibited. In comparison to CPS, the inhibitory rate ranged from 12% to 26% (gossypol treatment for 3 to 12 weeks, respectively). However, the synthetic activity for nuclear basic protein in the RPS was drastically reduced in comparison to that of the CPS. The reductive rate ranged from 46% to 61% in RPS (gossypol treatment for 3 to 12 weeks, respectively).Gel electrophoretic separation of the basic protein extracted from normal pachytene cell indicated that the major basic proteins were nucleosomal linker and core proteins, i. e., histone H_1, H_(2a), H_(2b), H_3 and H_4, and with a lesser amount of spermspecific BP and X_1 proteins. In the RPS, the synthesis of sperm-specific proteins(BP and X_1) and core histones (H_(2a) and H_4) reduced markedly.Finally, the effect of gossypol on the ratio change of nuclear basic protein to DNA in RPS and its relation to nucleosomal spacing and chromatin structure are discussed.
